For anyone needing it... This is the route I took, stay well clear of the "red" tiles
For one of the 4 Ankhs, the tile puzzle room

I carried one ankh from the first level (yes, I actually did). Does that mean I only need to find three ankhs here even though there are four?

I have a follow-up question regarding this particular puzzle. Originally I thought the openings in the ceiling corresponded to the ‘good’ tiles i. e. the ones that the player is supposed to step on or jump to, but apparently it’s the opposite: the openings indicate the ‘bad’ tiles, i. e. the ones that cause the door to close when you step on them (marked red in your map).

no not trial and error
it was obvious that the door closed when you touch
or jumped over a tile in the roof

But if you reached a safe tile, and you look for the next? You'll have to agree, very hard to see which one if you look up. You have to draw a map by counting the textures on the ceiling before you start and even that is quite difficult to do

So it’s like I thought: though the solution is not technically trial-and-error, in terms of gameplay it effectively is just that. I had thought about drawing a map myself, but I’m truly grateful that Dutchy did it before I had to. That puzzle was a real gameplay killer for me. I think the builder should have come up with a better way of indicating the bad tiles; and he easily could have, judging from the similar puzzle in the same level, where the good tiles are indicated by the feather of Maat.

Yeah you probably can, in the first level it was possible to get through the doors by only using one Ankh, so that one will stay in your inventory.
In level 2 there are 4 Ankhs needed, so with the extra one in your inventory you would only need to find 3
